Poster: Targeted in situ SNP genotyping for scalable human cell village studies
Population-scale studies using pooled human cell models are a powerful way to link genetic background to cellular phenotype, disease biology, and drug response, but they depend on reliable cell identity assignment. We demonstrate a targeted in situ SNP genotyping workflow to enable single-cell identity assignment within intact pooled cell models.
What you’ll learn:
- How a 115-SNP ID panel targeting high minor allele frequency SNPs in expressed genes was designed using universal human identification markers and GTEx data
- How the panel performed in across cell lines, individually and in defined mixtures, and how it scaled to a pool of genetically distinct cell lines
- How combining SNP identity, morphology, and gene expression can support cell village experiments, perturbation screens, genotype-to-phenotype mapping, and eQTL discovery.
