Poster: Spatially resolved mapping of intratumor clonal architecture using Direct In Sample Sequencing

Intratumor heterogeneity drives progression and therapeutic resistance, yet the spatial organization of subclones and how they interact with their local environment remains poorly understood. To advance cancer genomics research, we developed Direct In Sample Sequencing (DISS), a library prep-free workflow on the AVITI24™ platform for spatially resolved targeted RNA sequencing in intact tissue.

    What you’ll learn:

    • How DISS profiles up to 100 targets, covering known mutation hotspots in genes such as EGFR, KRAS, PIK3CA, and TP53.
    • How results compare to bulk RNA-Seq and targeted DNA sequencing of serial sections from the same tissue blocks.
    • How clonality data combines with high-plex RNA cell typing and 8-plex protein detection to classify immune-exclusion phenotypes around identified populations.
