Poster: Next generation multiplex hybrid capture and sequencing
Hybrid capture delivers deep, uniform coverage, but the workflow is complex and high-plex pooling can reduce library complexity diminishing sensitivity for low frequency variants. Here, we present a streamlined workflow using Trinity™ sequencing, paired with a new hybridization buffer that preserves library complexity.
What you’ll learn:
- How on-flow-cell capture and washing eliminate post-hybridization processing and post-hybridization PCR.
- How a novel hybridization buffer maintains high library complexity and uniformity with 24-sample multiplexing.
- Robust performance across exome and comprehensive genomic profiling (CGP) panels, including 87-88% on-target bases and 52-56x mean target coverage.
- How this workflow exhibited low duplication rates and high library complexity for sensitive variant detection.
