Please Note: We recommend users initiate the update during a period of instrument downtime. During the update process, the system may restart multiple times and will request a power cycle when the update is complete.
For systems with slower network connections and upgrading from AOS 2.x, the AOS 3.3.2 upgrade may take between 1-2 hours to complete. If the upgrade process does not complete within 3 hours, please contact ElemBio Support at support@elembio.com.
Summary of Changes
AVITI OS v3.3.2
AOS 3.3.2 contains improvements exclusively for AVITI24TM instruments and is an optional upgrade for AVITI TM instruments but is recommended for all AVITI systems not currently running AOS 3.3.1.
For AVITI24 instruments
- Resolved a known issue where reagent and buffer part numbers were hidden on the Run Summary screen during runs using a Custom Add-on Protein Panel
- Improved Polony Registration logic to capture Teton flow cell specific edge case scenarios
Known Issues, v3.3.2:
- If an upgrade to AOS3.3.2 does not complete within two hours, users should reach out to support@elembio.com for additional support
- If leak errors are triggered for both sides of the instrument, AVITI OS cannot unlock the waste module door.
- If the AFIC board loses power, it will not restart automatically and requires a power cycle. In the event of a power interruption or loss, power cycle the instrument via the UI after power is restored.
- The run can continue even if polony map generation fails, as AVITI OS cannot detect a particular failure scenario.
- The RunParameters.json Recipe Values field does not include the cartridge part number and small cell value.
AVITI OS v3.3.1
For all instruments
- Resolved a known issue where the Percent Passing Filter metric could be displayed prematurely, before the final calculation was complete.
- Resolved a known issue where "NaN%" was shown for the Q30 percentage of Library Pool 2 in dual library runs, instead of displaying a placeholder (e.g., "-") prior to the actual value populating.
For AVITI24™ instruments
- Resolved a known issue where in some cases, z-slices of Cell Paint tiles may be missing, causing the projected image for that tile to appear fainter.

AVITI OS v3.3.0
For all instruments:
- Performance improvements to the system have reduced the priming times required before each sequencing run. For instruments below high-altitude, priming time is reduced to less than 5 minutes. For high-altitude instruments, priming time is reduced to less than 8 minutes. Users should be aware of this reduced priming time and stay near the instrument to initiate the run.
- Over-the-air update improvements. Update procedure requires user to power cycle the instrument prior to upgrade, if a power cycle has not been performed in the prior week.
- Air Filter Replacement Reminder: For high-elevation sites, a reminder is sent every 6 months. For other sites, it is sent once a year. Users can replace the filter and reset the timer through AVITIOS.
- When an ElemBio Catalyst™ subscription expires
- Users will no longer be able to select the Catalyst storage connection during run setup.
- The storage connection will be marked as expired in the system.
For AVITI and AVITI LT instruments:
- AVITI and AVITI LT instruments now require a maintenance wash once every 30 days to maintain performance of the instrument. A reminder will be displayed to instrument users 60 hours prior to the maintenance wash deadline, to allow planning ahead.
For AVITI24™ instruments:
- Support for New Fixed Teton™ Immuno-Oncology and Neuroscience Panels
- New fixed Neuro and Immuno panels are now available in run setup.
- Includes new cell segmentation models for suspension and neuronal cell types.
- A small cell flag allows users to adjust amplification time for optimal signal detection.
- Support for New Teton Cartridge SKU
- Users can now select the cartridge directly from the run setup screen.
- The instrument UI has been updated to display the cartridge name throughout the workflow.
- The prepare reagents screen now reminds users to load RNA and protein probe tubes into the cartridge.
- The load reagents screen checks the cartridge part number against the selected cartridge.
- Support for Teton Custom Protein Add-On Panel
- Users may load a Teton Custom Add-On Protein Panel from USB or a published cloud design.
- The run setup screen now displays target names and validates panel content, ensuring:
- The panel is a B08 barcoding batch with 5-88 barcoding targets.
- The panel contains a valid panel name, ID, and probe concentrations.
- The instrument UI now displays the custom panel name in the run summary.
- When using an add-on custom panel, the prepare reagents screen prompts users to verify panel loading.
- OBPA execution now uses a merged panel that includes both the selected fixed panel and the additional batch.
