Which sequencing recipe should I use for my library insert size?

Introduction

Some NGS applications may target small or large molecules in the preparation method, resulting in insert sizes less than 100bp or more than 750bp long. Element provides sequencing recipes for specific insert size ranges that can improve the sequencing performance of these library sizes beyond the standard recipe. Recipe recommendations are shown in the table below. Please note that recipe recommendations are based on insert size, not library size.

Implementation

The short insert recipe and long insert sequencing recipes can be selected during manual run setup on the AVITI™ System user interface or planned run setup in ElemBio™ Cloud. During a Manual run setup, select Custom Recipe from the Advanced Settings menu. Then browse to either the short insert or long insert recipe in recipes list. For a planned run, specify the applicable recipe in the Advanced Run Settings section of the planned run form.

Recommendations

For complex or broadly distributed library pools, the standard recipe is recommended as a starting point. The short insert recipe can be used on a case-by-case basis for libraries with a significant population of small insert sizes that might be improved by the recipe.
