Which AVITI sequencing recipe should I use for my library insert size?
Answer
Most NGS libraries can be sequenced with the standard sequencing recipe on AVITI or AVITI24™. Some NGS applications target small or large molecules in the preparation method, resulting in insert sizes less than 100 bp or more than 750 bp long. Element provides sequencing recipes for specific insert size ranges that can improve the sequencing performance of these library sizes beyond the standard recipe.

Sequencing recipe recommendations are shown in the following table. Note that recipe recommendations are based on aligned insert size, not library size.

*The short insert recipe is required or recommended only for Cloudbreak Freestyle™. It is not needed for Cloudbreak™ or other applications.
Considerations
Implementation
The standard sequencing recipe is selected by default. You can select the short or long insert sequencing recipes during manual run setup on the AVITI or AVITI24 user interface, or during planned run setup in ElemBio™ Cloud.
During a manual run setup, select Custom Recipe from the Advanced Settings menu. Then browse to either the short insert or long insert recipe in the recipes list.
For a planned run in ElemBio Cloud, specify the applicable recipe in the Advanced Run Settings section of the planned run form.
Recommendations for Complex Pools
For complex or broadly distributed library pools, the standard recipe is recommended as a starting point. Use the short insert recipe on a case-by-case basis for libraries or pools with a significant population of small insert sizes that might be improved by the recipe.
