AVITI24 with OS v3.4.0 or later enables new computational improvements that enhance the performance of sequencing runs and increase pass-filter reads by up to 50% for many sequencing applications. Depending on the application, some libraries benefit from a slight loading increase to reach maximal output (up to 1.5 billion pass-filter reads) on AVITI24.

Comparison of AVITI with Expert Mode HD and AVITI24: scRNA-Seq Libraries
AVITI24 supersedes the gains from Expert Mode HD with most kit configurations (still available for 2x300 as this was not changed) while still maintaining high AVITI quality. For libraries that have previously benefited from the use of Expert Mode HD on AVITI, we’ve provided an example comparison of a scRNA-Seq library sequenced with standard settings on AVITI, with Expert Mode HD on AVITI, and with 1.25x loading on AVITI24.

Compared to a standard run on AVITI, Expert Mode HD provides ~20% more pass-filter reads for this scRNA-Seq library. AVITI24 provides ~338 million more pass-filter reads than AVITI with Expert Mode HD, or an increase of ~54% over the standard AVITI sequencing run for this library (Figure 1A). AVITI24 also maintains comparable quality, showing a slight increase over AVITI for this library (Figure 1B). Secondary analysis of these sequencing runs with Cell Ranger showed highly comparable metrics between all sequencing runs (Table 1). All runs also exceeded the Cell Ranger QC thresholds.
