Protein Overview
MAP2 is a dendrite-enriched microtubule-associated protein that cross-links and stabilizes microtubules, controlling dendrite formation, elongation, and neurite outgrowth. It also anchors PKA in dendrites, contributing to localized cAMP/PKA-CREB signaling important for synaptic plasticity.
Expected Localization
Somatodendritic - concentrated in neuronal cell bodies and dendrites, notably excluded from axons.
Immunofluorescence shows strong, relatively uniform staining of the dendritic arbor and soma with a sharp cutoff at the axon hillock.
Expected Expression Pattern
Expressed specifically in neurons of the CNS and PNS; used as a standard marker of neuronal identity and dendritic differentiation. Glial cells and non-neural cell types are essentially negative, though low-molecular-weight isoforms have occasionally been reported in reactive/cultured astrocytes.
Relevant Pathways
- Microtubule stabilization and dendritic cytoskeletal organization
- Dendrite outgrowth, branching, and arborization
- PKA anchoring and cAMP/PKA-CREB signal transduction
- Neurite polarity establishment (dendrite vs. axon)
- Synaptic plasticity-associated cytoskeletal remodeling

Qualitative Results
Localization Assessment
MAP2 protein panel demonstrates strong localization to microtubules (in iPSC-derived neurons and SH-SY5Y cells). The observed localization of MAP2 protein is consistent with published data.

iPSC-derived neurons were generated by pre-differentiating cells for 3 days in a 6-well plate, followed by transfer to Cytos Slide Kit. Cells were seeded at 1-3 K cells per well on Cytos Slide Kits, matured for 35 days with partial media changes every 3–7 days. After maturation, cells were fixed with 4% formaldehyde for 30 minutes, permeabilized using 70% ethanol for 10 minutes. The slide kit was then assembled into a flow cell and loaded onto an AVITI24™ along with the Cytos Protein Cartridge for a multiomics cell screening run. A549 and SH-SY5Y cells were also tested in parallel as part of Cytos cross-run reproducibility testing, where no detectable signal was demonstrated in the former. In iPSC-derived neurons and SH-Sy5y, MAP2 demonstrated strong signal intensity over negative controls with expected microtubule profiling and no detectable crosstalk. These results support high confidence in the low non-specific binding of this clone and validate this antibody as a passing clone.

- 8.1 ± 0.5 (SH-SY5Y; n=3 runs)
Indicates the fold brightness of the target signal as measured by the Mean Intensity Above (Otsu-defined) Threshold (IAT), over the background (a channel-matched isotype control) in the same cell. Data shown are mean ± standard deviation across independent reproducibility runs. The result of 8.11 in SH-SY5Y cells is over 4 times the acceptance threshold.

Panel-level run-to-run reproducibility |

For each imaging panel, we assess reproducibility by comparing the intensity signal a given antibody target produces across independent replicate runs. Within each run, we measure the average per-well fluorescence intensity for every target in the panel. We then plot each target's intensity from one run against the matching target's intensity from another run of the same layout, and calculate the Pearson correlation coefficient (r) between those two sets of measurements. Squaring that value gives the R² (coefficient of determination), which reports what fraction of the variation in one run's signal is explained by the other run's signal.
