What are my options for generating Elevate libraries?

The Elevate workflow is modular to allow you to mix and match library prep, enrichment, and index kits to configure your ideal prep.

The Elevate DNA Library Prep kit is available with either mechanical or enzymatic workflow options and can be paired with the Element Elevate Long UDI Adapter Kit Set A or Stubby Adapter UDI Primers for Element. You can also use Elevate adapter kits with third-party library prep kits that use A/T adapter ligation.

Summary

The modular nature of the Elevate workflow allows you to mix and match library prep, enrichment and index kits. Based on your experimental needs, consider the following combinations:

• Use Element Elevate DNA Library Kit and Long UDI Adapter plate for an end-to-end Elevate workflow with mechanical and enzymatic options for PCR-free or PCR-plus workflows
• Combine IDT Stubby Adapter UDI Primers for Element with a third-party library prep that uses A/T adapter ligation for PCR-plus workflows
• Combine Element Elevate Long UDI Adapter plate with a third-party library prep for PCR-free or PCR-plus workflows

Considerations

You can access detailed Elevate product information via the Elevate Workflow Product Page. Elevate libraries can be sequenced on AVITI™ and AVITI24™. Trinity™ and UltraQ™ sequencing require that input libraries have Elevate indexes and adapters. Element Elevate DNA Library Prep Kit and Elevate Long UDI Adapter Plate, Set A

These two products together make a complete whole genome sequencing (WGS) library prep kit. The Elevate DNA Library Prep Kit has options available for mechanically-sheared or enzymatically-fragmented libraries. Libraries can be prepared either PCR-free or PCR-plus.

Third-Party Library Prep Kit and IDT Stubby Adapter-UDI Primers for Element

Enables the use of Elevate Adapters with third-party library preps that use A/T adapter ligation. Libraries prepared with this workflow are PCR-plus. Transposase-based library preps are not compatible with this set. For more information, refer to the Element Elevate Library Prep User Guide for Third-Party Protocols.

Third-Party Library Prep Kit and Elevate Long UDI Adapter Plate, Set A

Enables the use of Elevate Adapters with a third-party library prep that is PCR-free with user-supplied surface primers and qPCR standards. To convert this workflow into a PCR-plus workflow, purchase oligos from a third-party supplier to generate amplification primers. Elevate Libraries for Trinity Sequencing

Trinity sequencing, which provides a simplified hybrid capture workflow, requires libraries prepared with Elevate indexes and adapters. Most hybrid capture workflows use the stubby adapter approach during library prep (see the Trinity Sequencing User Guide for more information). For a specialized PCR-free approach using a long UDI adapter set with Trinity, contact support@elembio.com for details and instructions for set-up. Trinity Freestyle™ is not compatible with Elevate indexes and adapters.
